Also look very carefully at the package you are on, both in terms of speed and cap, it's very easy to eat a lot of cap doing backups, and then get hit hard for expensive surcharges as a result.
The other thing to watch is that most broadband has a very slow upload, unless you are paying a lot for a very high end business package, so it might take a lot longer than you think to do the backup,
All the other comments are also very valid, and ideally, you need to be using 2 different physical locations alternately, so that if something goes wrong, you have still got a backup that is known to be good.
Also make sure that the remote site is making backups, so that you are not overwriting the only copy on their system.
Have fun, it's a nightmare trying to work out the best way to back up so much junk that may or may not be needed.
